Primary Reaction

ALT catalyzes:

L-Alanine + α-Ketoglutarate
→ Pyruvate + L-Glutamate

Indicator (Coupled) Reaction

Pyruvate + NADH + H⁺
→ Lactate + NAD⁺

As ALT activity increases, NADH decreases, and absorbance at 340 nm falls.
